客戶介紹:高杉,博士,教授,博士生導師,藥理學教研室主任,安徽醫科大學中青年學術技術帶頭人,美國University of Iowa訪問學者,新西蘭University of Otago短期訪問學者。研究方向:心血管藥理學。迄今指導畢業和在讀博士研究生4人,碩士研究生15人,其中6人榮獲國家研究生獎學金,2人榮獲優秀碩士論文。

實驗目的:提取肝臟組織DNARNA前期實驗室處理此類組織,主要是通過手持式研磨儀處理,單次處理量只有一個,耗時,耗力。又有處理樣本的批量比較大,處理的時間較長,樣品損失比較多,難以收集等缺點。現在實驗室這邊購買使用凈信研磨儀批量處理,速度快,重復性好,解決了實驗室目前樣品難處理的情況。

實驗地點、實驗材料:安徽醫科大學、肝臟組織若干,提取液,上海凈信冷凍研磨儀。
實驗步驟:
1.準備適配器,以及珠子分別放入離心管中。
2.加入珠子以及肝臟組織至研磨管。
3.將適配器裝入凈信JXSFTPRP-CL-24研磨儀中。
4.設定時間及參數,啟動儀器至研磨程序結束。
5、取出離心管,加入提取液,離心后取出研磨珠,回收后可反復利用。

得到如圖實驗樣本,經跑膠對比,實驗結果較實驗室手動處理,樣本較好!
注意事項:實驗準備階段提前五分鐘開啟冷凍研磨儀,腔體降溫,防止核酸降解。
實驗所用到的儀器簡單介紹以及后續所用設備:
研磨儀:批量處理肝臟組織樣本,研磨提取核酸。
離心機:離心提取上清液。
電泳槽:跑膠分離核酸,獲取目的條帶。
凝膠成像:拍照,用來得到實驗結果
