凈信多樣品組織研磨儀：樹枝內(nèi)微生物的研磨

　　研究方向：真菌多樣性與真菌病害
　　
　　實(shí)驗(yàn)?zāi)康模?/strong>根莖土壤內(nèi)微生物研磨，本次實(shí)驗(yàn)對象為微生物，要求與土壤一起磨，后續(xù)用來提RNA，沒用凈信研磨儀之前效果一直很差。
　　
　　實(shí)驗(yàn)步驟：
　　
　　1、準(zhǔn)備Tissuelyser-48一臺、鋼罐一套以及所需研磨樣品若干。
　　
　　2、取適量樣品、一顆研磨珠一起放入鋼罐中，蓋子旋上。
　　
　　3、將配備好的鋼罐放入液氮中預(yù)冷，凍脆樣品。（預(yù)冷5-10分鐘）
　　
　　4、將鋼罐適配器放入研磨儀中固定（適配器底部凹槽與軸的卡槽互相卡牢）放平后將鋼罐分別放入適配器對應(yīng)孔位，（注意平面配平，對稱放樣）蓋上適配器蓋，旋緊螺帽。
　　
　　5、設(shè)置參數(shù)（根據(jù)所需研磨量微調(diào)時間），開始研磨。
　　
　　6、磨好后打開儀器取出鋼罐，即可進(jìn)行后續(xù)實(shí)驗(yàn)。

　　上圖為1分鐘的研磨效果，已經(jīng)完全研磨完成。劉博士非常滿意，效果非常好，后續(xù)實(shí)驗(yàn)提出來的指數(shù)也非常好看。之前嘗試過多種前處理方式，人工研磨或者一系列粉碎機(jī)處理都非常費(fèi)時費(fèi)力，效果又很差，要是沒處理好時間久了rna還會降解，所以客戶經(jīng)過多次了解和對比所以老師們選用凈信研磨儀。效率高，效果好，完美解決了老師的研磨問題。
　　
　　注意事項(xiàng)：
　　
　　液氮操作需佩戴操作液氮的厚橡膠手套，注意安全。
　　
　　放鋼罐時時注意平面配平，只有一個樣品時需用空罐配平。
